Used For:
- Tacrolimus is an immunosuppressive medication used primarily to prevent organ rejection following transplant surgeries (kidney, liver, heart, etc.).
- It is also used in autoimmune conditions, especially in the topical form, to treat severe eczema (atopic dermatitis) and other skin conditions resistant to other treatments.
Side Effects:
- Common Side Effects: Headache, tremors, nausea, diarrhea, high blood pressure (hypertension), and difficulty sleeping.
- Serious Side Effects: Kidney damage (nephrotoxicity), liver dysfunction, infections (due to suppressed immune system), increased risk of cancer (especially skin cancer and lymphoma), high blood sugar (leading to diabetes), and neurotoxicity (e.g., seizures or confusion).
Precautions:
- Kidney Function: Tacrolimus can cause kidney damage. Renal function should be closely monitored with regular blood tests, especially in the early stages of treatment.
- Infections: As an immunosuppressant, tacrolimus increases the risk of infections, including opportunistic infections. Patients should avoid exposure to infections and follow infection prevention protocols.
- Blood Pressure and Blood Sugar: Tacrolimus may increase blood pressure and cause high blood sugar. Monitoring of these parameters is important during treatment.
- Pregnancy and Breastfeeding: Tacrolimus is classified as Category C during pregnancy, meaning it may harm the fetus. It should be used during pregnancy only if absolutely necessary, and it is excreted in breast milk, so caution is needed when breastfeeding.
- Drug Interactions: Tacrolimus has a high potential for drug interactions, especially with medications like azoles, macrolides, and anticonvulsants. Dose adjustments may be needed based on blood levels of tacrolimus and concurrent medications.
- Discontinuation: Stopping tacrolimus suddenly can lead to organ rejection, so it should be done under medical supervision.
